CellVia is a safe, sensitive colorimetric assay kit using a tetrazolium salt for the determination of cell viability in cell proliferation and cytotoxicity assays.
Recently, various kinds of tetrazolium salts(e.g. MTT, XTT, MTS, and so on) are available to measure cell proliferation or cell viability. These teterazolium salts are cleaved to formazan dye by the succinate-tetrazolium reductase, which exists in mitochondrial respiratory chain and is active only in viable cells.
CellVia uses a highly water-soluble tetrazolium salt, WST, which produces the water soluble formazan. The WST is reduced by dehydrogenase activities in cells to give a yellow-color formazan dye, which is soluble in the cell culture media. The amount of the formazan dye, generated by the activities of dehydrogenases in cells, is directly proportional to the number of living cells. The detection sensitivity of CellVia is higher than the other tetrazolium salts such as MTT, XTT, MTS or WST-1.
MTT - Limitations
MTT assay is a commonly used method among the cell proliferation assay using tetrazolium salt. In the method, MTT is reduced to a purple formazan by NADH. However, MTT formazan is insoluble in water, and it forms purple needle shaped crystals in the cells. Therefore, prior to measuring the absorbance, an organic solvent is required to solubilize the crystals. Additionally, the cytotoxicity of MTT formazan makes it difficult to remove cell culture media from the plate wells due to floating cells with MTT formazan needles, giving significant well-to-well error.
More sensitive and convenient tetrazolium salt has been required because of the inconveniences above.
As a result, Water Soluble tetrazolium salts such as MTS, XTT, WST have become a new standard for Cell Viability Assay instead of MTT.
New Water Soluble Tetrazolium Salts
Water Soluble tetrazolium salt solves all disadvantages of MTT assay as indicated above.
